Antimicrobial activity of topical agents against Propionibacterium acnes: an in vitro study of clinical

This study aimed to compare the antimicrobial activities of topical agents against Propionibacterium acnes isolated from patients admitted to a hospital in Shanghai, China. The minimal inhibitory concentrations of the cultured P. acnes were determined in accordance with the Clinical and Laboratory Standards Institute. Susceptibilities to clindamycin and erythromycin were compared in terms of gender, age, disease duration, previous treatment, and disease severity. A total of 69 P. acnes strains were isolated from 98 patients (70.41%). The susceptibility to triple antibiotic ointment (neomycin/bacitracin/polymyxin B) and bacitracin was 100%. The susceptibility to fusidic acid was 92.7%. The resistance rates to neomycin sulfate, erythromycin, and clindamycin were 11.7%, 49.3%, and 33.4%, respectively. The high resistance rate to clindamycin and erythromycin was significantly affected by gender, previous treatment, and disease severity rather than by age and disease duration. Topical antibiotics should not be used separately for long-term therapy to avoid multiresistance. The use of topical antibiotics should be determined by clinicians on the basis of clinical conditions.

摘要: A novel hybrid material, Cu-PAA/MWCNTs (copper nanoparticles deposited multiwalled carbon nanotubes with poly (acrylic acid) as dispersant, was prepared and expected to obtain a more effective and well-dispersed disinfection material for water treatment. X-ray energy dispersive spectroscopy (EDS), transmission electron microscopy (TEM), the X-ray fluorescence (XRF), X-ray photoelectron spectra (XPS), Fourier transform infrared spectra (FT-IR), Raman spectroscopy, and thermal gravimetric analyzer (TGA) were used to characterize the Cu-PAA/MWCNTs. ( ) was employed as the target bacteria. The cell viability determination and fluorescence imaging results demonstrated that Cu-PAA/MWCNTs possessed strong antimicrobial ability on . The deposited Cu was suggested to play an important role in the antimicrobial action of Cu-PAA/MWCNTs.

多重耐药菌的出现迫切需要发现具有新作用机制的抗生素。在本研究中,笔者将疏水的吖啶分子连接到核定位序列 (NLS) 的N末端合成了一种新型的抗菌药物Acr3-NLS。为了进 一步提高该试剂的抗菌活性,笔者将两个单体Acr3-NLS分子通过二硫键连接合成了二聚体(Acr3-NLS)2。结果显示,与NLS相比,Acr3-NLS,特别是(Acr3-NLS)2,对革兰阴性菌和革兰阳性菌表现出显著的抗菌活性。随后,其作用机制的研究表明,Acr3-NLS 和(Acr3-NLS)2 可通过细胞膜破坏方式和DNA 结合方式杀死细菌。作用于细胞膜和细胞内DNA的双靶点抑杀机制可以降低细菌对Acr3-NLS 和(Acr3-NLS)2 产生耐药性的风险。总之, 本研究提供了一种新的策略,可用于设计具有双重作用机制的高效抗菌药物。

Antimicrobial use in food animal production: situation analysis and contributing factors

摘要:

By measuring the quantity and ways in which antimicrobials are used, and reviewing different technical and socioeconomic factors influencing antimicrobial use at farm level, this study discusses the main knowledge gaps in antimicrobial use in food animal production and provides recommendations for future research and policy development. The review reveals that antimicrobial use in food animals exhibit strong regional and species differences, and there are still large information gaps concerning the current state of antimicrobial use. Factors associated with animal health (including antimicrobial resistance), animal health improvement, economic costs and benefits relevant to animal diseases, and potential technological alternatives or alternative systems all have an impact on antimicrobial use on the farm. There is a clear need to resolve the data gap by monitoring antimicrobial use and developing an analytical framework to better understand farmer behaviors under different technical, economic and environmental circumstances.

Discovery of novel ursolic acid derivatives as effective antimicrobial agents through a ROS-mediated

摘要: In response to the reduction of food production and economic losses caused by plant bacterial diseases, it is necessary to develop new, efficient, and green pesticides. Natural products are rich and sustainable source for the development of new pesticides due to their low toxicity, easy degradation, and eco-friendliness. In this study, we prepared three series of ursolic acid derivatives and assessed their antibacterial ability. Most target compounds exhibited outstanding antibacterial activities. Among them, the relative optimal EC50 values of Xanthomonas oryzae pv. oryzae and Xanthomonas axonopodis pv. citri were 2.23 (A17) and 1.39 (A16) μg·mL–1, respectively. The antimicrobial mechanism showed that compound A17 induced an excessive accumulation and production of reactive oxygen species in bacteria and damaged the cell membrane integrity to kill bacteria. More interestingly, the addition of low concentrations of exogenous hydrogen peroxide enhanced the antibacterial efficacy of compound A17 against Xanthomonas oryzae pv. oryzae. These entertaining results suggested that compound A17 induced an apparent apoptotic behavior in the tested bacteria. Overall, we developed the promising antimicrobial agents that destroyed the redox system of phytopathogenic bacteria, further demonstrating the unprecedented potential of ursolic acid for agricultural applications.

Antifungal and antibacterial functions of medicinal leech recombinant destabilase-lysozyme and its heated-up derivative

摘要: Antifungal activity of recombinant medicinal leech destabilase-lysozyme (rec.Dest-Lys) was investigated by using fungi: , and , including yeasts and . Its antibacterial activity was investigated on gram-negative bacteria . These activities were assessed by radial agar diffusion assay, and scanning and transmission electron microscopy. Therefore, destabilase-lysozyme not only is endo-isopeptidase and lysozyme, but also has antifungal and antibacterial activities. Muramidase activity of rec.Dest-Lys disappeared after heat-treating at 90°C for 50 min without the loss of its antimicrobial activity. Furthermore we showed for the first time that the heated-up derivative of rec.Dest-Lys exhibited more potent activities against the above enumerated fungi and gram-negative bacteria than original protein.

摘要: 2-aryl-3-(naphthalene-1 or 2-yl)-1,3-thiazolidin-4-ones and were synthesized in 41%-67% yield by using microwave-assisted one-pot protocol. The structures of the new compounds , , , , , , and - were confirmed by IR, NMR, MS, and elemental analysis. The antimicrobial activities of the compounds against pv. lachrymans (Smith et Bryan) Young, Dye & Wilkie, Pers., and Blum. were examined. Some of the compounds showed good antifungical activity against Blum.

越南广平和顺化两省猪场分离副猪嗜血杆菌的抗生素耐药表型和耐药基因之间的相关性研究 Article

副猪嗜血杆菌是一种对全球养猪业造成重大经济损失的病原菌。副猪嗜血杆菌防控失败的原因之一是细菌耐药性不断加重。越南拥有亚洲排名第二的生猪产量,但是缺乏针对副猪嗜血杆菌耐药性的相关研究和数据。本研究的目的是调查越南副猪嗜血杆菌的耐药性,并分析耐药性和耐药性基因之间的关系。本研究所使用的副猪嗜血杆菌分离菌株来自我们之前研究中从越南广平省和顺化省猪场分离到的菌株。本研究分别利用肉汤微稀释法和聚合酶链反应(PCR)法检测了这些菌株对25种抗生素的耐药性及相关耐药基因的存在情况。结果显示,这些分离株对甲氧苄啶/磺胺甲恶唑的耐药率达高到94.6%,其次是黏菌素、氯霉素、庆大霉素、青霉素、林可霉素和阿莫西林。这些菌株中相关联耐药基因的检出率分别为blaTEM-1 94.6%、int 76.8%、gyrA 58.9%以及rmtD 50.0%。头孢呋辛、氯霉素和妥布霉素的耐药性表型分别与耐药基因blaROB-1 [优势比(OR) = 26.3, 95% 置信区间(CI)2.7~255.7, p = 0.002]、catl (OR = 25.1, 95% CI 2.4~258.9, p = 0.004)以及strB (OR = 23.5, 95% CI 2.6~212.6, p = 0.001)呈显著正相关。本研究首次揭示了越南中部地区副猪嗜血杆菌的耐药性情况,有助于该疾病的临床控制,能够为降低越南中部的养猪业中该菌的耐药性以及为相关政策制定和临床用药提供指导。

IDEEA activity monitor: validity of activity recognition for lying, reclining, sitting and standing

Recent evidence demonstrates the independent negative effects of sedentary behavior on health, but there are few objective measures of sedentary behavior. Most instruments measure physical activity and are not validated as measures of sedentary behavior. The purpose of this study was to evaluate the validity of the IDEEA system’s measures of sedentary and low-intensity physical activities: lying, reclining, sitting and standing. Thirty subjects, 14 men and 16 women, aged 23 to 77 years, body mass index (BMI) between 18 to 34 kg/m2, participated in the study. IDEEA measures were compared to direct observation for 27 activities: 10 lying in bed, 3 lying on a sofa, 1 reclining in a lawn chair, 10 sitting and 3 standing. Two measures are reported, the percentage of activities accurately identified and the percentage of monitored time that was accurately labeled by the IDEEA system for all subjects. A total of 91.6% of all observed activities were accurately identified and 92.4% of the total monitored time was accurately labeled. The IDEEA system did not accurately differentiate between lying and reclining so the two activities were combined for calculating accuracy. Using this approach the IDEEA system accurately identified 96% of sitting activities for a total of 97% of the monitored sitting time, 99% and 99% for standing, 87% and 88% for lying in bed, 87% and 88% for lying on the sofa, and 83% and 83% for reclining on a lawn chair. We conclude that the IDEEA system accurately recognizes sitting and standing positions, but it is less accurate in identifying lying and reclining positions. We recommend combining the lying and reclining activities to improve accuracy. The IDEEA system enables researchers to monitor lying, reclining, sitting and standing with a reasonable level of accuracy and has the potential to advance the science of sedentary behaviors and low-intensity physical activities.

Production of pectic extracts from sugar beet pulp with antiproliferative activity on a breast cancer

摘要: In the last years, sugar beet pectins have been the subject of several investigations involving extraction methodologies, chemical composition and functional properties. The structure of pectins, which depends on the extraction method, is decisive in their capacity to induce apoptosis on several cancer cell lines like colon, prostate and breast. In this work, sugar beet pectin extraction was performed in the following steps: lipid extraction with hexane, removal of soluble complex carbohydrates and proteins, and enzymatic treatment with amyloglucosidase, protease, and pectinase. The enzymatic treatment was carried out with Rohapect DA6L under the following conditions: 50°C, pH 4.0, 2% enzyme/substrate (E/S) ratio, 15 h, and a solid to liquid ratio of 1 ∶ 10. The pectic extract showed a degree of polymerization (DP) profile of 55.8% with DP≥7; 4.9% with DP6; 5.8% between DP2 and DP6 ; 4.7% with DP2; and 28.8% with DP1. The pectic extract was examined for its antiproliferative activity on the MCF-7 breast cancer cell line. At a concentration range of 12.5–25 mg/mL the pectic extract killed 80.6% of the cells, exhibiting a higher antiproliferative activity than 4-hydroxytamoxifen (4-OHT), a classical anticancer drug, which killed 56.5% of the cells.

Strategies on designing multifunctional surfaces to prevent biofilm formation? ?

摘要: Bacteria adhesion and biofilm formation have raised severe problems on public health, food industry and many other areas. A variety of reagents and surface coatings have been developed to kill bacteria and/or limit their interaction with surfaces. It has also attracted many efforts to integrate different bactericidal elements together and maximize antibacterial efficiency. Herein, we review mechanisms for both passive and active approaches to resist and kill bacteria respectively, and discuss integrated strategies based on these two approaches. We also offer perspective on future research direction.

Low intensity ultrasound stimulates biological activity of aerobic activated sludge

摘要: This work aims to explore a procedure to improve biological wastewater treatment efficiency using low intensity ultrasound. The aerobic activated sludge from a municipal wastewater treatment plant was used as the experimental material. Oxygen uptake rate (OUR) of the activated sludge (AS) was determined to indicate the changes of AS activity stimulated by ultrasound at 35 kHz for 0 40 min with ultrasonic intensities of 0 1.2 W/cm. The highest OUR was observed at the ultrasonic intensity of 0.3 W/cm and an irradiation period of 10 min; more than 15% increase was achieved immediately after sonication. More significantly, the AS activity stimulated by ultrasound could last 24 h after sonication, and the AS activity achieved its peak value within 8 h after sonication, or nearly 100% higher than the initial level after sonication. Therefore, to improve the wastewater treatment efficiency of bioreactors, ultrasound with an intensity of 0.3 W/cm could be employed to irradiate a part of the AS in the bioreactor for 10 min every 8 h.

preparation of Ag@AgCl/Bio-veins composites and their photocatalytic activity and recyclability

摘要: The industrial application of nano-photocatalysts in wastewater treatment has been severely restricted for a long time due to their difficult separation, poor reusability, and low efficiency. In this work, a facile strategy was proposed to enhance the photocatalytic activity and recovery performance of Ag@AgCl nanocatalysts. Biological veins (Bio-veins) with a unique 3D porous construction were used as carriers for the in-situ growth of Ag@AgCl nanoparticles. Scanning electron microscopy results showed that the Ag@AgCl nanoparticles were uniformly loaded on the surface and interior of the Bio-veins, and the size of the Ag@AgCl nanoparticles immobilized on the Bio-veins (50–300 nm) was significantly smaller than Ag@AgCl obtained by the co-precipitation method (1–3 μm). The Bio-veins played a vital role in the photocatalysis reaction system. The degradation efficiency of the Ag@AgCl/Bio-veins(CI4) was up to 3.50 times as high as pure Ag@AgCl. Furthermore, the composites also exhibited excellent recyclability and stability under both visible and solar light. This work provided a suitable strategy for nano-photocatalysts for practical application and may also offer new possibilities for the high-value utilization of biomass materials.
